Mam problem z Mod Rewrite Friendly URLs, gdy ustawię ten parametr i przejdę do jakiegoś tematu otrzymuje błąd 500,

Błąd 500 występuje po stronie serwera.
Dlaczego zdublowałeś wpisy {REQUEST_FILENAME} dwa razy?

Proponuję wybadać która linijka powoduje błąd.
Zacząłbym od zakomentowania RewriteBase.
